APVV-20-0241 Role of the circadian system in colorectal cancer progression mediated by estradiol and miRNA. 1.7.2021 – 30.6.2025, principal investigator

We successfully performed screening of rhythmic miRNA in the rat colon and determined their relationship with the presence of rhythmic miRNA precursor. We revealed surprising clustering of miRNA acrophases at the beginning of dark phase of light:dark cycle which is in accordance with higher expression of DGCR8 (component of miRNA biosynthesis pathway). By extensive in silico research, we determined PTEN and p53 regulatory pathways as cross points in the circadian and miRNA mediated regulation in CRC. Reasons for sex dependent CRC incidence were also considered in this respect.

APVV-16-0209 Circadian and ontogenetic regulation of miRNA in mammals. 1.7.2017 – 30.6.2021, principal investigator.

The most important output of this project is analysis of the daily profile in expression of key components of miRNA biosynthesis: Drosha, DGCR8 (DiGeorge syndrome critical region 8) and Dicer. Both enzymes and cofactor DGCR8 were determined in the central oscillator (suprachiasmatic nuclei of the hypothalamus) and in the peripheral oscillators (heart, liver, and kidneys) during a 24h cycle in animals synchronized under light(L):dark(D) conditions 12L:12D.

VEGA 1/0455/23 Generation of rhythmic pattern in miRNA expression. 1.1.2023 – 31.12.2026, principal investigator

Project is focused on the role of miRNA in colorectal cancer treatment progression and treatment. Potential use of miRNAs is also approached. In particular we analyse way how specific tissues contribute to miRNA spectrum in the circulation. We are interested in miRNA interference with long ncRNA and regulation of miRNA levels by sponging, degradation and export from cells and/or tissues.

VEGA 1/0679/19 The role of clock genes and functionally related miRNAs in the execution effects of factor regulating cell cycle progression. 1.1.2019 – 31.12.2022, principal investigator.

Among the most important outputs of the project belongs discovery of interaction between miR-34a-5p with the circadian system, as miR-34a-5p significantly inhibits per2 expression and stimulates expression of clock. We found that colorectal carcinoma cell line DLD1 intensively expresses mRNA ESR2, while ESR1 mRNA is present at a low level, or it is under detection limit. While ESR2 expression is not affected by miR-34a-5p, the expression of ESR2 mRNA is significantly inhibited by miR-150-5p. This finding is consistent with in silico analysis (Target Scan Human, Release 7.2). Using a wound healing assay, we demonstrated that miR-150-5p in tumour cell line DLD1 stimulates cell migration, which is in accordance with decreased ESR2 mRNA expression.

APVV-14-0318 microRNA analysis and characterization of expression of selected proteins in circadian context as a prognostic biomarker for colorectal cancer 1.7.2015 – 30.6.2019, investigator.

The project included miRNA screening to identify miRNAs that show the most significant differences in expression between tumor and healthy tissues, and simultaneously, are well measurable in human plasma samples of patients diagnosed with colorectal cancer.
