Key outcomes
Booster immunisation against diphtheria in adults carried out with bivalent tetanus-diphtheria vaccines produced a protective immune response dependent on persistent levels of diphtheria antibodies prior to the booster dose. The persistence of antibody levels is the key predictor of seroprotection achievement correlating with time since the last vaccination.
Therefore, two-dose vaccination should be considered, in an emergency, in recipients with the last dose received >34 years earlier. Based on our data, we can further anticipate a higher increase in diphtheria antibodies more often in non-smokers and in adults without statin treatment. If these factors have any clinical impact on protection against diphtheria is yet to be established.

Key outcomes
Our research highlighted the fragmentation of food supplement regulation at the EMS level regarding the responsible authorities’ legislative frameworks and regulatory practices. It also endorsed the claim that the functionality of mutual recognition is dependent on EMS acceptance and illustrated that mutual recognition of food supplements is not accepted by EMS and hence not functional as a governance mode in practice. We assume that the fragmentation of the regulation of food supplements in the EU is associated with the dysfunctionality of mutual recognition of food supplements at the EU level. Strengthening the harmonization of food supplement regulation could enhance access to safe food supplements and the free movement of goods throughout the EU. Improved secondary legislation on product safety databases or communication procedures between authorities should be considered. Initiatives such as the BELFRIT project demonstrate that this is explicitly desired by EMS. Continued research in this area, with a focus on the involvement of other EU countries and an inclusion of new participants, e.g., business operators, food supplement quality and safety laboratories, consumers, and others, could initiate a deep revision and acceleration of the food supplement harmonization by all relevant stakeholders.

Key outcomes
Intervention of pharmacists in medication adherence can meaningfully contribute to achieving therapeutic outcomes. Exploring the real-life readiness and opportunities of pharmacists may result in the adoption of measures, which could be seen through improvement of patients ́ adherence to pharmacotherapy.
The pharmaceutical care is developing towards the improvement of medication adherence in both countries, the Czech Republic and Slovakia, assuming a medium degree of adherence support. Further progress may be observed in strengthening the pharmacists’ personnel capacities, and accelerated mainly using information technologies.

As part of experimental research, growth factor-like substances associated with MHV-68, named MHGF-68, were discovered in our laboratory. MHGF-68 activity was manifested by the ability to alter cell morphology, that is, normal phenotype to transformed, resp. suppresses the transformed phenotype of tumour cells. The aim of the experiments was to monitor the effect of MHGF-68 on the change of the cell actin cytoskeleton in the tumour cell line Hepa1c1c7, as well as the normal cell line NIH3T3, and compare conventional stationary cultivation and dynamic cultivation conditions using a LiveFlow system (In Vitro Technologies). LiveFlow is an advanced system to test the impact of different compounds on the cell cultures, which allows simulation of in vivo conditions thanks to continuous flow of cultivation medium. MHGF-68 was prepared with the infection of BHK-21 cells with MHV-68 virus under non-permissive conditions (41°C). After dynamic cultivation with MHGF-68, we observed changes in morphology on Hepa1c1c7 cells. In cells cultured in a dynamic environment, we observed more pronounced changes in cell morphology in comparison with cells cultured statically. We observed no changes in the cytoskeletal structures in the NIH 3T3 cell line affected by MHGF-68 in both types of cultivation. The advantage of LiveFlow in comparison to in vivo testing is that the experiments performed in this system are less time and money consuming. Dynamic cultivation in the LiveFlow system is suitable for optimizing experiments before testing substances in vivo.
