Project Summary:


Natural compounds have always attracted interest due to their potential effects on human being. Their use is widespread and is rather rising due to false belief that they are always better and safer than conventional drugs. This belief is often the background for companies producing preparations containing such compounds. Hence, the major aim of this project is to prepare comprehensible, free and easily accessible materials reporting both positive and negative aspects of natural compounds used for several commonly used indications (infections, menopause and gynaecological problems, benign prostatic hyperplasia, cough, cardiovascular diseases, CNS, skin and GIT disorders), as well as of vitamins and sympathomimetics, according to the most recent scientific evidence. Special materials will be also prepared for their interactions with conventional drugs.


The novelty and strength of this material will be based on several aspects:

1. They will be freely available,

2. There will be three versions, one for professionals in the form of open access publications, one for students of biomedical disciplines and one for laypersons, both latter as attractive e-learning material in Moodle platform. In particular, in the current situation in the World, the need for a quality and easily accessible e-learning materials is apparent.

3. Materials for students and laypeople will be prepared in English and 8 native languages of participating universities (Czech, French, German, Hungarian, Italian, Portuguese, Slovak and Slovene),

4. Materials will be prepared by common work of experts from different fields (pharmacologists, pharmacognostics, toxicologists, microbiologists, analytical chemists and E-learning experts).


The team is well balanced not only from the scientific point of view, but it also encompasses collegues with solid experiences in a similar e-learning project (e.g. TOX-OER project, www.toxoer.com) as well as many young and talented colleagues.

Summary:

Rheumatoid cachexia leads to muscle weakness, and loss of functional capacity, and is thought to accelerate morbidity and mortality in rheumatoid arthritis (RA). Adjuvant arthritis (AA) is a model of RA that involves cachexia and also leads to changes in body composition similar to those of RA patients. Our project will examine the relationship between basic arthritic parameters, inflammation, oxidative stress, lipid profile, hunger hormones and satiety. For this purpose, we use AA induced in Lewis rats. Three different plant extracts from olive leaves, Rhodiola rosea, Tribulus terrestris and virgin olive oil alone and in combination with methotrexate are used to reduce cachexia, inflammation and oxidative stress. These plant extracts and virgin olive oil have several biological effects (anti-inflammatory, antioxidant, adaptogenic, anabolic, etc.) and may be beneficial as adjunctive therapy to standard antirheumatic drugs in reducing arthritic cachexia.

Summary:

The project aims to reveal the unknown mechanisms of etiopathogenesis of RA, up to the level of expression of important Nrf2 and NF-kappa B genes involved in the regulation of inflammation and redox balance of the organism. For this purpose, we will use an experimental model of RA, namely adjuvant arthritis (AA) induced in Lewis rats. We will characterize the development of AA with parameters describing immunological, oxidative and inflammatory processes. In this model, in combination with the acute model (carrageenan-induced inflammation) and cell culture (BV-2 mouse microglia stimulated with inflammation), we will also evaluate the antirheumatic effect of substances not previously studied in adjuvant arthritis in rats (antioxidants: astaxanthin, troloxolocarnoz selected phytochemicals and extracts, punic acid, Lactobacillus plantarum strain LS / 07). The most effective substances of the individual groups are then selected in combination with a basal antirheumatic drug - methotrexate. The expected benefit is an increase in the efficacy of methotrexate without increasing the risk of side effects.
