VEGA 1/0203/19 Targeted downregulation of endothelin-1 in pulmonary arterial hypertension


Project summary:

We plan to contribute new knowledge on the pharmacological modulation of endothelin-1 (ET-1) expression in

pulmonary arterial hypertension(PAH). The strong vasoconstrictor and mitogenic peptide ET-1 plays a key role in

the pathogenesis of this fatal disease. Current therapies aim at blocking the action of ET-1 or at dilating the

pulmonary artery acting on the smooth muscle. Hitherto unused modulation of ET-1 expression could become an

attractive therapeutic target. In monocrotaline model, we shall determine effects of small molecules (that we

previously found to inhibit ET-1 expression in cultured vascular smooth muscle cells) on morbidity, mortality,

hemodynamics and ET-1 expression In cultured vascular endothelial and smooth muscle cells, we shall study

stable antisense and siRNA oligos directed against ET-1 mRNA acting on posttranscriptional level, with the aim to

find optimal structures usable in an in vivo experiment to degrade ET-1 mRNA regardless of the stimulus that

increased ET-1 gene transcription.
