VEGA 2/0429/21Study of mechanisms of modulation of inflammation and lipid metabolism by lactobacilli in a model of non-alcoholic fatty liver disease - co-investigator.
The project aims to study the mechanisms of the effect of 4 lactobacilli strains (i) on the inflammatory process and lipid metabolism in a liver model (HepG2) and (ii) on the disrupted intestinal barrier (Caco-2) in vitro. In the first stage, we will focus on the study of the TLR4 signaling pathway and its downstream molecules, on the activation of anti-inflammatory cytokine production, and on the stimulation of the expression of negative regulators of the TLR4 signaling pathway in the liver. We will monitor the influence of lactobacilli on the regulation of lipid metabolism (influx, efflux, synthesis, and degradation of cholesterol, triacylgycerols, fatty acids). In the second stage, we optimize the inflammatory coculture model of enterocytes (Caco-2) with hepatocytes (HepG2). At the molecular level, we will analyze the ability of lactobacilli to restore the impaired permeability of the Caco-2 monolayer and affect the inflammatory process and lipid metabolism in a model of interorgan communication over time. With our knowledge, we would like to contribute to the elucidation of the mechanisms of NAFLD and the possibilities of its influence by probiotics.
VEGA 1/0394/18 Phosphatidylinositol system and transcription factors in the context of secondary metabolites - co-principal investigator
The project focused on the study of phosphatidylinositol-4-phosphate-5-kinase (PI4P-5K, EC 2.7.1.67) and WRKY / MYB transcription factors in the context of signaling systems and plant secondary metabolism. Poppyseed (Papaver somniferum L.) is used as the experimental model. The effect of phospholipid signaling on the production and accumulation of plant secondary metabolites has not been thoroughly investigated. So the question is: Can the phosphatidylinositol system affect the biosynthesis of benzylisoquinoline alkaloids? And what are the functions of transcription factors in these processes? The aim of the project is to identify the complete cDNA sequence encoding poppy PI4P-5K, to monitor the transcriptional level WRKY / MYB, to determine the profile of alkaloids (morphine, codeine, thebaine, papaverine) and gene transcripts for alkaloid biosynthesis enzymes (TYDC-OMT, 4-T, OMT, SalAT, COR).
KEGA 064UK-4/2018 Innovation of Immunology and Microbiology Practical Classes and Elaboration of Textbook – co-investigator
The project aimed to develop updated university textbooks for practical exercises in the compulsory subjects of Immunology and Microbiology for students of the Pharmacy study program. These subjects will bind the modern and actual knowledge and changes in pharmaceutical or medical teaching processes. The project also included selecting and introducing modern methodologies suitable for implementation in university laboratories. The updating and preparation of textbooks concerned not only their content but also their form of concept. The specific output is two new textbooks: Immunological Methods: principles and instructions for practical exercises by Kiňová Sepová H., Bilková A., Hrčka Dubničková M., Dudík B. (FaF UK, Bratislava 2021) and Practical exercises in microbiology for pharmacists by Drobná E., Hrčka Dubničková M., Greifová G. (FaF UK, Bratislava 2021), which are available to all students of the Faculty of Pharmacy, Comenius University in Bratislava, both prepared in online form. In addition to instructions, the textbooks also contain theoretical introductions to individual tasks, which the students will find application in the practical part of learning the given subjects. They also will be the material suitable for self-study during the preparation for final exams in these subjects.

In cooperation with the Radiology Laboratory of the JINR in April, the topic of Research of Biological Action of Heavy Charged Particles with Different Energy no. 04-9-1077-2015 / 23 (Biological reactions to low dose radiation). the effect of radiation on apoptosis and necrosis of immunocompetent cells after lipid A treatment was studied. In these cases, the observed DNA damage after radiation and regeneration of diphosphoryl- and monophosphoryl lipid A cells in vivo was continued. The alkalic version of the gel electrophoresis - comet assay (Comet assay) was used in vitro to detect single- and double-stranded DNA breaks at the leukocyte level. Real in vitro results showed that the highest number of DNA breaks was observed in irradiation alone samples that correlated with diphosphoryl lipid A-regenerated cells. The action of gamma radiation (60Co) is planned further, the results are compared with each other and the hypothesis of the protective ability of MLA on individual cell types will be described in terms of a statistically significant result of the project. The whole intention is made from the intention of protecting the cells of the immune system and the hippocampus from the limb system of the human brain, which plays an important role in consolidating information from short-term memory to memory and especially spatial orientation in conditions of ionizing radiation.
