APVVV-16-0600 Štruktúra a reaktivita aptamérov a vybraných molekulových systémov v

patologickej forme priónov /Structure and reactivity of aptamers and selected molecular

systems in the pathological form of prions / 7/2017 - 12/2021

Poznanie molekulových interakcií je kľúčom pre pochopenie mechanizmov procesov

prebiehajúcich v živých organizmoch. Predkladaný projekt navrhuje analýzu interakcií

RNA/DNA aptamérov a vybraných molekúl so sekvenciami priónov N-domény, určenie

štruktúry ktoré je stredobodom záujmu výskumu, pomocou MD,

kvantovomechanických výpočtov a UVVIS spektrofotometrie. Výsledky aplikácie teoretických

metód umožnia analyzovať termodynamické a štruktúrne parametre komplexov priónov s

vybranými molekulovými systémami. / Knowledge of molecular interactions is a key concept in

the understanding of the biological mechanisms of life.The given project proposes the analysis

of the interactions of RNA/DNA aptamers as well as selected molecules with the peptide

sequences of prions of the N-domain by using of MD, quantum-chemical calculations, UVVIS

spectrophotometry. The results from the application of theoretical methods allow to analyse

thermodynamical and structural parameters of the complexes of prion with selected molecules

and to find the binding site of molecules at the prions. The project looks for the conditions

leading to the increase of the ligand activity to prions. The study connects in the case of

radical analysis also the experimental research with the application of theoretical methods.
